Should the red line of the IDE be where I've marked the black arrow?

Yes, it's pin 1, but the problem I think you have based on your photo you seem to have your PC power supply plugged into the GW. That socket is shown as power OUT, not in.
That plug on the GW is to power your floppy not to plug power in from your PC. The FDD doesn't get power over the data cable, so you need to unplug floppy power from the GW and hope you didn't kill it.
Just plug FDD power into the FDD itself and USB and the floppy cable into the GW.
The other thing here is that the GW is just a USB device you use under windows using the software from the GW github. It isn't detected like a normal floppy and won't do anything until you instruct it to do so.
That plug on the GW is to power your floppy not to plug power in from your PC. The FDD doesn't get power over the data cable, so you need to unplug floppy power from the GW and hope you didn't kill it.

I took the power out of the GW but now there is no power plug in as I only have this open ended cable.
I ran the software and nothing happens.

Plug that power cable into the GW and into your floppy power connection. Plug the 34 pin cable into the floppy and GW. Plug the GW into USB and it should all power on.
If your FDD needs 12v though, you will have to feed it power from your PC as the GW can only do 5v. Also, the USB you are plugged into may not have enough power for the GW and FDD, sou you'd be better with a separate PSU for the FDD, i.e. direct from your PCs PSU.

Should be 5V.
